You will work on a project that aims to unravel important mechanisms that contribute to inflammatory bowel disease in humans. Work is based on our previous findings that monocytes and macrophages in the intestine of patients with active inflammatory bowel disease have diminished expression of a key cell surface receptor called integrin αvβ8, which is a crucial activator of the anti-inflammatory cytokine TGFβ. Our preliminary data now suggest that soluble mediators are responsible for downregulation of this pathway in the inflamed intestine, and this project now aims to characterise the molecules and mechanisms responsible.
The project will involve liaison with gastroenterologists in local hospitals, acquisition of human blood and tissue samples, and analysis of cells and molecules by flow cytometry and other analytical techniques. The project aims to provide a step-change in our understanding of how monocytes and macrophages become dysregulated in patients with inflammatory bowel disease.
