Cerebral small vessel disease (SVD) is a leading cause of vascular cognitive impairment and dementia. Despite the importance of SVD there are few treatments for established disease, a situation hindered by a lack of understanding of disease mechanisms. A unique grouping of clinical and pre-clinical expertise in SVD, matrix, vascular biology and inflammation from five major UK universities including University of Edinburgh, University of Glasgow, University of Cambridge, University of Manchester and University of Oxford, have assembled to work together, forming a consortium (www.vadmatrix.com), aiming to identify key molecular mechanisms of SVD, which is critically important to identify potential therapeutic targets.
In the central nervous system, small blood vessels and neuronal cells are tightly integrated forming a dynamic multicellular structure, called the neurovascular unit (NVU). Complex and dynamic interactions between different types of cells in the NVU regulate the blood-brain barrier and the blood flow in the brain, shaping brain function in both health and disease. Central to maintaining NVU integrity is the extracellular matrix (ECM), together with associated proteins, forming the matrisome. This project is designed to use patient-specific induced pluripotent stem cell (iPSC) models, together with state-of-art technologies including bioimaging, functional analysis of NVU, proteomics and transcriptomics, to decode the function of the matrisome in SVD.
The project will be based in the School of Biological Sciences, FBMH, led by Professor Tao Wang, in collaboration with Professor Stuart Allan, as part of a multi-University consortium (www.vadmatrix.com).
The applicant should have a PhD degree in the area of biology or biomedical related subjects. He/she will have a demonstrated research experience in cell biology and molecular biology. Experiences in stem cell biology or vascular and/or neural cell biology are desirable.
